Ipamorelin is a synthetic pentapeptide growth hormone secretagogue developed by Novo Nordisk in the late 1990s. Structurally, it belongs to the GHRP family but stands apart from earlier members (GHRP-2, GHRP-6, Hexarelin) through a critical distinction documented in peer-reviewed comparisons: high selectivity. At equimolar doses, Ipamorelin stimulates GH release in preclinical models without the corresponding cortisol, ACTH, or prolactin elevations observed with other GHRPs—making it the cleanest tool available for isolated GH-axis research.
Ipamorelin acts as an agonist at the ghrelin receptor (GHS-R1a), stimulating GH secretion through a pathway distinct from and complementary to GHRH. This is why combining Ipamorelin with a GHRH analog (such as CJC-1295 Without DAC) produces synergistic GH release in animal models—the two compounds work through mechanistically independent but convergent signaling pathways at the pituitary somatotroph. Published pharmacokinetic studies show a half-life of approximately 2 hours in rats, consistent with a clean pulse of GH release.
