IGF-1 LR3 is an engineered analog of native insulin-like growth factor-1, modified at two levels to substantially extend its biological activity window in research systems. First, position 3 carries an arginine substitution (R³) that dramatically reduces affinity for IGF-binding proteins (IGFBPs), which in native IGF-1 sequester up to 99% of circulating peptide and restrict its receptor access. Second, a 13-amino acid N-terminal extension peptide is prepended to the mature IGF-1 sequence, totaling 83 amino acids compared to the 70 of native IGF-1.
The consequence of IGFBP avoidance is significant in cell culture and preclinical models: IGF-1 LR3 remains receptor-available at far higher effective concentrations than native IGF-1 at equal doses, and with a reported half-life approximately 60-fold longer in preclinical studies. This makes it the preferred IGF-1 variant for in-vitro proliferation and signaling studies, where it activates the IGF-1R→PI3K/AKT→mTOR and MAPK/ERK pathways with a prolonged and clean signal. It is a standard tool in cell biology and muscle biology research for precisely this reason.
