What a triple agonist is

Most incretin-class peptides studied to date act on one or two receptors. Retatrutide is structurally notable because a single molecule carries agonist activity at three.

The incretin system is a set of gut-derived hormone signals that participate in glucose regulation and energy metabolism. Research peptides in this class are designed to bind and activate specific receptors in that system. A mono-agonist targets one receptor; a dual agonist targets two. Retatrutide is designed as a triagonist — one peptide chain with activity at three distinct receptor targets.

That matters to researchers for a practical reason: comparing a triagonist against mono- and dual-agonist compounds isolates what the third receptor contributes, which is difficult to study by combining separate molecules with different pharmacokinetics.

Receptor targets

Target 01

GLP-1 receptor

Glucagon-like peptide-1 receptor. The most extensively characterised incretin target, and the basis of the single-agonist peptide class that preceded this one.

Target 02

GIP receptor

Glucose-dependent insulinotropic polypeptide receptor. The second incretin axis, and the addition that defines the dual-agonist generation of research compounds.

Target 03

Glucagon receptor

The third axis, and the one that distinguishes a triagonist. Glucagon receptor signalling sits on the opposing side of glucose regulation from the incretin receptors, which is why combined activity in a single molecule is of structural interest.

Published literature

Retatrutide is an investigational compound. It has not been approved by the FDA or any comparable regulator for any use. The following peer-reviewed publications are provided so researchers can locate the primary literature directly. They are cited as literature — APEX 100 Research makes no claim about outcomes, efficacy or suitability for any purpose.

Common questions

What does “GLP-3 RTA” refer to?

GLP-3 RTA is our catalogue designation for this research material. The compound is retatrutide, development code LY3437943. CAS 2381089-83-2.

What does ≥99% HPLC purity mean?

High-performance liquid chromatography separates the sample into its components and measures each as a proportion of the total. ≥99% means the target peptide accounts for at least 99% of the material by that method. The chromatogram showing this is included in the lot’s COA.

Can I see the COA before ordering?

Yes. Sample documentation is on our COA page, and we will send the specific COA for any lot on request — before or after purchase.

Is retatrutide an approved drug?

No. Retatrutide is investigational. It has not been approved by the FDA or any comparable regulator for any use. It is supplied here strictly as a research material.

How is this shipped?

Sourced and shipped within the United States, typically 2-day delivery. Lyophilized peptides are stable in transit; refrigerate on arrival.

What is the difference between the 10 mg and 30 mg vials?

Mass of lyophilized compound per vial. Both are the same material to the same ≥99% specification, with the same documentation. The 30 mg vial carries a lower cost per milligram.

Analytical verification

Every lot is tested before release and ships with its own certificate of analysis. We publish the COA unredacted — including the lot number, the test date and the actual chromatogram — rather than a summary figure.

HPLC purity

High-performance liquid chromatography establishes purity against the ≥99% specification. The chromatogram is included in the COA, not just the resulting number.

Mass spectrometry

Identity is confirmed by mass spectrometry against the expected molecular weight of 4731.33 g/mol.

Lot traceability

Each vial’s lot number corresponds to a specific COA on file. Request the document for any lot you have received at any time.

Handling and storage

Supplied lyophilized. Standard laboratory handling for peptides of this class applies.

Lyophilized storage36–46 °F short term; −4 °F for extended storage, protected from light
ReconstitutionBacteriostatic water is the common choice for laboratory reconstitution. Add solvent slowly against the vial wall; do not inject directly onto the powder.
MixingSwirl gently until dissolved. Do not shake or vortex — mechanical agitation can shear peptide chains.
After reconstitutionRefrigerate at 36–46 °F. Avoid repeated freeze-thaw cycles.
LightProtect from light at all stages.
